Previous studies have obtained mammalian haploid embryonic stem cells, but haploid embryonic stem cells undergo spontaneous diploidization during in vitro culture and differentiation, posing challenges for the establishment of haploid somatic cell genetic screening systems. Zhou Qi, a member of the Chinese Academy of Sciences and a researcher at the Institute of Zoology, Chinese Academy of Sciences, confirmed by live cell observation that mitotic slippage of haploid embryonic stem cells during division causes cells to enter the interphase directly from the medium term, resulting in diploidization. Screening with small molecule inhibitors that regulate key targets in the mid-stage of division revealed that the CDK1 and ROCK pathways are key pathways regulating the diploidization of haploid embryonic stem cells. By adding a ROCK inhibitor, haploid embryonic stem cells can be differentiated into haploid cells of three germ layers, including neural stem cells (which can be further differentiated into mature neurons and astrocytes), cardiomyocytes, and islet cells. The haploid neuron mutation library constructed using the PiggyBac transposon system can be used for the screening of the resistance gene of neurotoxin Mn2+, and it is confirmed that Park2 plays an important role in the neuronal resistance to Mn2+ neurotoxicity. The above work revealed the occurrence of haploid diploidization and found that the CDK1 and ROCK pathways are critical pathways for regulating the process of diploidization.
